Establishment of Highly Efficient Plant Regeneration, Callus Transformation and Analysis of Botrytis cinerea-Responsive PR Promoters in Lilium brownii var. viridulum

Lilium brownii var. viridulum, commonly called Longya lily, is a well-known flower and vegetable plant in China that has poor tolerance to Botrytis fungal disease. The molecularimprovement mainly been restricted an efficient regeneration transformation system. In this study, the highly of lily was established through optimization embryogenic callus, adventitious shoot rooting induction. major factors influencing (antibiotics, Agrobacterium concentration, infection time, suspension solution coculture medium) were examined. expression responses PR promoters (ZmPR4 BjCHI1) B. cinerea assessed transgenic calli. results showed Murashige Skoog (MS) medium with 1.0 mg·L−1 picloram (PIC) 0.2 1-naphthaleneacetic acid (NAA) under light conditions MS 0.5 6-benzylaminopurine (6-BA) NAA darkness optimal for callus induction (64.67% rate) proliferation (3.96 coefficient). Callus inoculation into containing 2.0 thidiazuron (TDZ), 0.4 NAA, TDZ led shooting (92.22 (3.28 coefficient) promotion, respectively. rate reached 99.00% on 0.3 NAA. Moreover, 65.56% achieved by soaking at OD600 10 min modified without NH4NO3 as before selecting 75 hygromycin 300 cefotaxime. Only BjCHI1 promoter obviously expressed These could facilitate generation plants improved resistance.
